嗜酸粒细胞促淋巴细胞活性及抗肿瘤的作用机制 被引量:1

Antitumor and immunoregulation effects of eosinophils in vitro

摘要 目的:探讨嗜酸粒细胞(EOS)体外抗肿瘤及对淋巴细胞活性的影响,研究EOS在肿瘤免疫中的作用.方法:建立豚鼠I型超敏反应模型,应用Percoll液不连续密度梯度法分离豚鼠血液、腹腔液中EOS.放射免疫法检测豚鼠血清及EOS与淋巴细胞共同培养的上清液(SELC)中TNF-α,IL-4含量;MTT法分别测定EOS,SELC,EOS裂解液(LLE)对白血病K562细胞,乳腺癌MCF-7细胞增殖活性的影响.结果:豚鼠血液、腹腔液EOS(1×106,1×107,1×108/L)能够促进脾淋巴细胞增殖,刺激指数最高分别可达1.92,2.58.血清,SELC中IL-4,TNF-α含量明显增高(P<0.05);腹腔液SELC48hIL-4,TNF-α含量最高分别为(3.94±0.16),(8.93±0.15)g/L;腹腔液EOS,SELC,LLE与K562,MCF-7细胞共同培养,能够抑制K562,MCF-7细胞的增殖,尤其SELC对K562增殖的抑制率最高可达59.28%.结论:EOS可以促进淋巴细胞的增殖活性,提高SELC中IL-4,TNF-α的含量.腹腔液EOS,SELC,LLE能够抑制肿瘤细胞的增殖. AIM: To explore the antitumor effect of eosinophils (EOS) on K562 and MCF-7 ceils and the immumoregulation activity in lymphocytes. METHODS: Type I hypersensitivity model was established in Guinea pigs and eosinophils was isolaied from the peripheral blood and peritoneal fluid. Cytokines( TNF-α, IL-4) in blood serum and in supernatant by coculture of eosinophil and lymphoeytes (SELC) were measured with radioimmunoassay, and the proliferation of K562 and MCF-7 cells treated with EOS, SELC and lysis liquid of eosinophil(LLE) was respectively assessed by tetrazolium salt reduetion(MTT) assay. RESULTS: The lymphocyte proliferation from the spleens in guinea pig was promoted with peripheral blood EOS and peritoneal fluid EOS( 1 × 10^6, 1 × 10^7 , 1 × 10^8/L) , and the proliferation index was up to 1.92 and 2.58 respectively. The levels of TNF-α and IL-4 in SELC significantly increased ( P 〈 0.05 ) and up to ( 3.94 ± 0.16) and (8.93 ±0.15) g/L respectively. The proliferation of K562 and MCF-7 cells was respectively inhibited by peritoneal fluid EOS, SELC and LLE and the inhibition rate in the growth of K562 cells with SELC was the highest( 59.28% ). CONCLUSION: EOS promotes the lymphocytes proliferation activity and enhances the levels of TNF-α and IL-4 in SELC. The peritoneal fluid EOS, SELC, LLE inhibits the activity in tumor cells in vitro.
