摘要
目的探讨茶多酚对高糖条件下大鼠晶状体上皮细胞形态及生长情况的影响。方法取第2代对数生长期的大鼠晶状体上皮细胞,给予不同浓度的葡萄糖、茶多酚进行培育24小时,观察高糖条件下及茶多酚干预后大鼠晶状体上皮细胞形态的变化,MTT法检测0.1μM、0.3μM浓度的茶多酚对高糖培养的大鼠晶状体上皮细胞生长情况的影响。结果在高葡萄糖浓度的培养条件下,大鼠晶状体上皮细胞死亡率增加,生长受到抑制。而用茶多酚干预的高糖条件的大鼠晶状体上皮细胞则较好的保持了上皮细胞的形态,细胞死亡较少,生长良好。大鼠晶状体上皮细胞的存活率和抑制率两两比较差异有统计学意义(P<0.05),不同浓度的茶多酚两组之间比较差异无统计学意义(P>0.05)。结论茶多酚对高糖损伤的晶状体上皮细胞具有保护作用。
OBJECTIVE To study the effect of TP on the morphology and growing of rat lens epithelium in high-glucose. METHODS The second generation rat's lens epithelium cells were cultured in glucose and TP of different concentration for 24h. The morphous changes of the cells were then observed when they were intervened in different concentration of TP. The effect of 0.1μM, 0.3μM TP to the growing of rat lens epithelium in high-glucose was determined by MTT assay. RESULTS Rat lens epithelium cells'death rate increased and growing was inhibited in high-glucose. Few rat lens epithelium cells died in high-glucose which was intervened by TP, their growing was good and the morphous was not destroyed. There was a significant difference between the survival rate and inhibition rate (P〈0.05), there was no significant difference between the TP of different concentration (P〉0.05). CONCLUSIONS The morphous of rat lens epithelium cells were abnormal, the amount of cell decreased in high-glucose; the morphological changes of the lens epithelium cells and amount's decrease were delayed and prevented after intervened by TP,maintained lens epithelium cells's inherent characteristics,showing that TP could protect lens epithelium cells from being damaged by high-glucose,establish the base for the study of whether or not TP could prevent or cure diabetic cataract and the causes.
