短期和长期间歇运动对大鼠心肌和冠状血管肾上腺髓质素的影响 被引量:9

Effect of Short-term and Long-term Interval Exercise on Adrenomedulin in the Myocardium and Coronary Vessels of Rats

摘要 目的:研究3天和3周间歇运动对大鼠心肌和冠状血管肾上腺髓质素(Adrenomedullin,ADM)的影响,探讨短期和长期间歇运动对心肌和冠状血管保护作用的机制。方法:25只SD大鼠,参照文献建立短期和长期间歇运动动物模型,以30m/min的速度在跑台上运动15min、休息5min,重复3次。分别以这种方式连续运动3天(3D组,n=12)和3周(3W组,n=13),另取12只大鼠不运动作为对照组(C组)。采用酶联免疫法检测大鼠血清和心肌ADM含量,免疫组织化学法检测心肌和冠状血管ADM表达,并应用计算机图像分析技术对心肌和冠状血管阳性表达面积和平均光密度进行定量分析。结果:与C组比较,3D组和3W组血清ADM含量变化不明显,3D组心肌ADM含量变化不明显,3W组心肌ADM含量明显升高(P<0.05);与C组比较,3D组心肌和冠状血管ADM表达无明显变化,3W组心肌ADM表达显著增强。计算机图像分析结果显示,3W组心肌ADM阳性表达面积和平均光密度明显高于C组(P<0.05),冠状血管阳性表达面积和平均光密度明显低于C组(P<0.05)。结论:长期间歇运动可增加心肌ADM分泌,可能有利于维持血液动力学稳定和保护心肌。 Objective To probe the cardioprotection mechanism of short-term and long-term interval exercise,the effects of 3-day and 3-week interval exercises on the adrenomedulin (ADM)in myocardium and coronary vessels of rats were studied. Methods Twenty five adult male Sprague-Dawley rats were forced to run on a treadmill at the speed of 30m/min intermittently,three sections per day,5 minutes per section,with an interval of 5 min rest. The exercise in short-term group(3D)lasted for 3 days(n=12)and in long-term group(3W)lasted for 3 weeks(n=13); other 12 rats were served as control group(C). The content and expression of ADM were detected with euzymelinked immunosorbent assay and immunohistochemical technique. The immunoreaction area and average optical density were analyzed quantitatively through computer image analysis. Results As compared with the group C:(1)the content of serum ADM changed insignificantly both in groups 3W and 3D;(2)the content of myocardial ADM increased significantly in group 3W(P 〈0.05),while changed insignificantly in group 3D;(3)the expression of ADM in myocardium and coronary vessels in group 3D changed insignificantly,while the expression of ADM in myocardium in group 3W changed significantly;(4)the immunoreaction area and mean optical density of myocardial ADM in group 3W increased significantly(P 〈0.05),and the immunoreaction area in coronary vessels decreased significantly (P 〈0.05). Conclusion The results suggested that long-term interval exercise increases secretion of ADM in myocardium of rats,playing an influential role in maintaining the stability of blood dynamics and in protecting myocardium.
