摘要
对黑曲霉AS3.4309液体曲产糖化酶的培养条件进行了研究。采用糖化力较强的黑曲霉进行液态培养,在单因素试验的基础上,采用4因素3水平正交试验对其产糖化酶的培养条件进行优化。结果表明,该菌株产糖化酶的最优培养基组成为:玉米粉15%,玉米浆7%,麸皮5%,豆粕粉2%,(NH4)2SO40.15%;培养条件为:接种量10%,发酵液起始pH4.5,30℃,200r/min,摇瓶培养7d,最高酶活力达到1294.52U/g。
The liquid fermentation conditions for the glucoamylase production by Aspergillus niger AS 3.4309 were studied. Based on single factor test, the glucoamylase production conditions were optimized by L9 (3^4) orthogonal test. The results indicated that the optimal medium contained 15% corn starch, 7% com steep liquor, 5% bran, 2% soybean flour and 0.15% (NH4)2SO4. The culture conditions were as follows: inoculum size 10%, ini- tial pH 4.5, temperature 30℃, shaker speed 200 r/min and culture time 7 d. Under above conditions, the highest glucoamylase activity reached 1294.52 U/g.
