摘要
目的:探讨中药连翘对大肠杆菌R质粒的消除作用,使其重新恢复对抗生素的敏感性。方法:将大肠杆菌DH5α制备成感受态后,导入含有Ampr基因的质粒,再用连翘作用转化后的大肠杆菌,经培养12h后,挑出亚抑菌浓度的菌落涂于含有Amp的平板上培养24h,观察有无细菌生长,并通过电泳检测是否消除质粒。结果:在含有Amp平板上无大肠杆菌生长,在不含Amp的平板上细菌生长良好,质粒DNA电泳检测结果经中药作用过的大肠杆菌无质粒带,表明连翘对质粒具有消除作用。
objective： Study on Chinese traditional medicine Weeping Forsythia to eliminate R plasmids of E. coll. , thus to restore the sensitivity to antibiotics. Methods： After the E. coli. was made of competence imported, the plasmid which contained Ampr ,then cultivated the E. coli. was transformed by the Weeping Forsythia, after cultivating 12h, picked out the subinhibitory concentration colony to overlay the plate, cultivate 24h, then observe whether the E. coli. grew. Result： The E. coli. was not found on the plates with Amp, but be found on the plates without Amp, the E. coli which was effected by Chinese traditional medicine didn＇ t have plasmid, it indicated that the Weeping Forsythia can eliminate the plasmid of the E. coli.
