摘要
目的探讨茎叶人参皂甙(GS)对内毒素(LPS)诱导的急性肺损伤大鼠模型肺组织中白细胞介素-8(IL-8)表达的影响。方法SD大鼠30只随机分为正常对照组(n=10)、内毒素损伤组(n=10)、茎叶人参皂甙组(n=10)。给药后5 h,观察各组大鼠的一般状况,取动脉血1 mL行动脉血气分析后处死大鼠,肉眼观察肺组织改变,取左肺计算肺湿重、肺干重及湿干重比,取右肺作病理学检查,免疫组化法检测各组肺组织IL-8的表达,病理图像分析系统测定肺组织中的平均灰度值。结果与正常对照组比较,内毒素损伤组大鼠一般状况较差,肉眼观肺组织水肿出血明显,肺W/D升高,动脉血PaO2显著下降,病理学检查肺组织损伤明显,肺组织IL-8表达显著升高;而茎叶人参皂甙组与内毒素损伤组比较一般状况较好,肉眼观察肺组织水肿出血不明显,肺W/D降低、动脉血PaO2下降不明显,病理学检查肺组织损伤较轻,肺组织IL-8表达轻度升高。结论茎叶人参皂甙可以通过部分抑制肺组织IL-8的表达,起到减轻肺损伤的作用。
Objective To study the effect of Stem and Leaf Ginsenoside on expression of interleukin-8 in LPS induced acute lung injury albino rats' lung. Methods 30 SD albino rats were randomly divided into: normal group ( n = 10) , LPS model group ( n = 10) , GS group ( n = 10) ,5 hours after administration, observing the rats' condition, taking blood out of abdominal aorta and doing blood gas analysis immediately, then putting mice to death, observing lung issue by eye, weighing the left lung wet weights, dry weights, rate of wet weights to dry weights of this rat. observe right lung issue by visual inspection. The expression of IL-8 in the lung tissue was detected by immunhistochemical method and pathology image analysis system determination IL-8 average grey. Results GS model group' s condition was worse than normal group but better than LPS model group,pulmonary edema: GS model group was worse than normal group,better than LPS model group, normal group' s lung W/D are less than other groups, LPS model group' s lung W/D are more than GS group ; normal group' s PaO2 was bighter than other groups, GS group' s PaO2 was highter than LPS model group;the results of pathology: normal group was batter than other groups, LPS model group was less than GS group; The expression of IL-8 of normal group was the least in all of groups, The expression of IL-8 of LPS group was more remarkable, The expression of IL-8 of GS group was slightly. Conclusion Stem and Leaf Ginsenoside can reduce lung injury by partly inhibiting the expressions of inflammatory factor as IL-8.
