尿激酶RGDS靶向溶栓造影剂的制备及鉴定 被引量:7

摘要 目的探索制备同时携带尿激酶及精氨酸-甘氨酸-天冬氨酸-丝氨酸(RGDS)靶向溶栓造影剂的可行性,并评价其理化性质。方法尿激酶、RGDS荧光双标记后,根据尿激酶及RGDS不同质量比(1∶1、2∶1及1∶2)分为3组,采用直接连接法制备,并检测三组微泡大小、形态、荧光亮度,微泡与尿激酶及RGDS的结合率。结果通过直接连接法可以将声诺维、尿激酶及RGDS三者结合,以尿激酶和RGDS呈1∶1比例配制的最佳,洗涤前、后微泡表面均可激发出绿色及橙红色荧光,流式细胞仪检测结果显示,洗涤后尿激酶携带率为(72.3±9.4)%,RGDS携带率为(64.6±10.2)%。结论携带尿激酶及RGDS的靶向溶栓造影剂能够成功制备。 Objective To prepare a thrombus-targeted ultrasound contrast agent carrying Urokinase (UK) and Arg-Gly- Asp-Ser (RGDS) peptides, and to assess its physiochemical properties. Methods After labeling UK and RGDS in different fluorescence, contrast agent was prepared using a direct connection method. This study was divided into three groups according to different ratio of UK/RGDS. The size, shape, fluorescent intensity, the binding rate of UK and RGDS were measured and analyzed. Results SonoVue, UK and RGDS were combined using direct connection method. The contrast a- gent of preparation with UK/RGDS 1:1 was optimum. The surface of prepared microbubbles emitted green and oranged fluorescence under the fluorescent microscope. The Flow cytometry test showed the binding rate of UK after washing was of (72.3±9.4) %, while the binding rate of RGDS was of (64.6 ± 10. 2)%. Conclusion The thrombus-targeted ultrasound contrast agent carrying UK and RGDS can be prepared successfully with the above-mentioned method.
