摘要 实验建立了沙门氏菌的磁捕获-PCR检测方法。筛选了最佳包被液和最适包被条件,用于制备包被有抗沙门氏菌抗体的磁珠。用制备的免疫磁珠捕获样品中的沙门氏菌,用于PCR检测。此方法操作简便,用时短,检出率高。用所建立的免疫磁珠捕获-PCR技术检测牛乳中的沙门氏菌,检测灵敏度为9cfu/mL,所用时间为7h(包括前增菌时间)。 In order to develop a rapid detection method of Salmonella in milk product, magnetic particles coated with the antibody specific for Salmonella was used to separate Salmonella from spiked milk followed by DNA release and PCR amplification. Different immunomagnetic beads and different coated solution were evaluated an.d selected. And a quick IMB-PCR detect system was established. The detection limit is 9 cfu/mL in 7 hours.
