CTGF mRNA在糖尿病大鼠心肌中的表达及葛根素和氨基胍的调节作用 被引量:3

摘要 目的:观察结缔组织生长因子(CTGF)在糖尿病大鼠心肌组织中的表达情况及葛根素(Pue)、氨基胍(AG)对CTGF表达的调节作用。方法:给大鼠腹腔注射链脲佐菌素(STZ)制备糖尿病模型,用葛根素(Pue1组,160mg·kg-1·d-1;Pue2组,120mg·kg-1·d-1;Pue3组,80mg·kg-1·d-1)、氨基胍(AG组,100mg·kg-1·d-1)治疗12周;采用RT-PCR检测心肌组织CTGFmRNA的表达,透射电镜下观察心肌细胞的形态学改变。结果:糖尿病组大鼠心肌组织CTGFmRNA的表达升高,Pue2组、Pue3组和AG组CTGFmRNA的表达明显降低;电镜下可见糖尿病组大鼠心肌细胞肌原纤维含量明显减少,治疗组大鼠心肌病变明显减轻。结论:葛根素(120和80mg·kg-1·d-1)和氨基胍能下调CTGFmRNA的表达,对糖尿病大鼠心肌具有保护作用。 Objective: To observe the mRNA expression of the connective tissue growth factor(CTGF)in myocardial tissue in rats with diabetes mellitus (DM) and the adjusling effects of puerarin (Pue) and aminoguantdine (AG) on CTGF expression. Methods: The diabetes mellitus (DM) rats models were made by intraperitoneal injection of streptozotocin (60 mg/kg body weight), then treated with Pue and AG. After 12 weeks, the rats were sacrificed. The expression of CTGF mRNA of myocardial tissue were determined with reverse transcription polymerase chain reaction (RT-PCR). Morphology of myocardium of rats were also observed under transmission electron microscope (TEM). Results: The CTGF mRNA expression was significantly higher in DM group than those in NC group and Pue treated (120 mg·kg^-1·d^-1,80 mg·kg^-1·d^-1)and AG treated groups (P〈0.01). It showed that myofibrilla contents in myocardial cell decreased significantly under TEM in diabetic group. The pathological changes of myocardium which induced by diabetes were reversed in different doses of Pue treated groups and AG treated groups. Conclusion: The protective effect of Pue and AG on myocardium of diabetic rat may be down-regulated through decreasing the level of CTGF mRNA exnression.
