摘要
目的:观察阿魏酸对兔血管平滑肌细胞骨架F-actin的作用。方法:兔血管平滑肌细胞用含20%灭活胎牛血清的高糖DMEM培养,在阿魏酸作用24h后,用鬼笔环肽标记细胞骨架蛋白F-actin后,用流式细胞仪检测荧光强度,用激光共聚焦显微镜观察细胞骨架蛋白的变化。结果:阿魏酸作用24h后,流式细胞检测结果显示,细胞骨架蛋白F-actin荧光强度明显减弱,与对照组比较,有显著性差异。激光共聚焦显微镜观察可见细胞形态明显变长,荧光减弱,与对照组比较,荧光密度减弱。结论:阿魏酸对兔血管平滑肌骨架蛋白F-actin的形成有抑制作用。
Objective: To observe the effect of ferulaic acid on the F-actin cytoskeleton of rabbit vascular smooth muscle cells (VSMCs). Methods: VSMCs were cultured with high glucose DMEM containing 20% deactivated fetal bovine serum, the cells were treated with ferulaic acid for 24 h and then F-actin was labeled with FITC-phalloidin, and the fluorescence intensity was detected with FCM, and then observed with confocal microscopy. Results: FCM result showed that F-actin of VSMCs was decreased after treated with ferulaic acid for 24 h. The fluorescence intensity of F-actin was obviously weakened, and it showed a significant difference compared with control group. The confocal microscopic observation showed that the outlines of F-actin of VSMCs were longer, and the fluorescence was weaker, and the fluorescence density was also weaker compared with control group. Conclusion: Ferulaic acid can inhibit F-actin formation of VSMCs.
