摘要
为研究共固定化细胞因子(肿瘤坏死因子-α+干扰素-γ)诱导HeLa细胞凋亡的有效性及长效性,并初步探讨其中的Fas死亡受体信号转导途径,本实验利用光化学固定法制备接枝细胞因子的高分子药物,在不同剂量(20 ng/well和200 ng/well)、不同时间(1、3和6 d)条件下诱导细胞凋亡,流式细胞术检测HeLa细胞凋亡过程中Fas基因的表达量。结果表明,游离细胞因子具有较快的诱导HeLa细胞凋亡活性,共固定化细胞因子具有更好的长效诱导凋亡活性。Fas死亡受体信号可能参与了固定化细胞因子药物诱导HeLa细胞凋亡。
This study was aimed to examine the expression of apoptosis-associated gene Fas in HeLa cell, explore the effects of the co-immobilized cytokines (tumor necrosis faetor-α and interferon-γ), and probe the potential mechanism of action. The preparation and application of the researeh couple IFN-γand TNF-α to the polystyrene cell culture plate were performed using the Photo-immobilization method, with different doses (20 ng/well and 200 ng/ well) and synthesized optical active material. HeLa cells were treated with eytokines for two dose and 1, 3, 6 days. The result showed that the free eytokines indueed HeLa apoptosis quiekly, yet the HeLa apoptosis induced by co-im- mobilized cytokines had longer effeet.
