摘要
以可溶性淀粉和海藻酸钠混合胶为固定化载体,戊二醛为交联剂,通过固定化乙醇氧化酶(AOX)制成核微孔酶膜,构建出过氧化氢电极系统。同时研究了该酶电极的特性,以及温度、海藻酸钠浓度、pH等相关参数对酶膜检测能力的影响。实验结果表明,在pH8,35℃时酶膜活性最佳,当乙醇体积分数为0.01%~1%时响应值有良好的线性关系,最低乙醇检测浓度在0.001%以下,响应时间为20s。通过对酶膜活性半衰期的检测,建立了乙醇氧化酶型酶膜优化工艺条件,其性能良好,具有较好的应用前景,特别适用于乙醇的在线监测。
The research constructed an ethanol sensors by building nuclear pore film, which is made by AOX immobilization. The system contained the immobilized carrier via mixing Soluble Starch and Sodium Alginate, and chose Glutaraldehyde to be the cross-linking agent. The research also tested the specialty of enzyme electrode,temperature, coneentration of sodium alginate, pH and concerning parameter which influence the detecting ability of memb'rane. The results showed that the membrane have the best.detection ability in the atmosphere of pH8 and 35℃. When the alcohol concentration ranges from 0.01%to 1% the response value has a good linear relationship. The lowest detectable concentration is less than 0. 001%, and the responding time is 20s. Finally, we found the optimum production technology of alcohol oxidase enzyme membrane, by means of testing the half time of membrane. It must be a wonderful application prospect because of the high performance and being suitable for online detecting.
