摘要
为了探讨肿瘤化疗机理,作者采用流式细胞术单参数分析方法,检测了45例恶性肿瘤患者化疗前后外周血细胞的凋亡(Apo)、S期细胞比率(SPF)和增殖指数(PI)的变化。结果表明,肿瘤患者血细胞Apo水平明显高于正常对照(P<0.01),化疗结束时进一步升高(P<0.01),化疗后1个月又恢复到化疗前的水平。肿瘤患者的SPF和PI均显著高于正常对照(P<0.01),化疗结束时明显减低,PI尤为显著(P<0.01),并持续到化疗后1个月。从而可见,化疗使患者血细胞Apo呈“一过性”增高,肿瘤的化疗机制主要是化疗药物使细胞增殖活性减低,细胞聚积在Go/G1期,同时药物诱导细胞凋亡增加。
In order to investigate the effective mechanism of anti tumor chemotherapy,Apo (apoptosis),SPF (S phase fraction) and PI (proliferation index) in PBM (peripheral blood monocytes) of 45 patients with malignant tumor were measured by flow cytometry. The results showed that the Apo levels in patients group were higher than that in normal control ( P <0.01),They were further increased after chemotherapy ( P <0.01) and at one month later returned to the previous level. Also, SPF and PI,in especial PI,in patients group were higher than that in normal control ( P <0.01). They were reduced after chemotherapy ( P < 0.01 ) and the reduced levels continued for at least one month. The data indicate that the cellular apoptosis and inhibited cellular proliferation may be a main mechanism of anti tumor chemotherapy.
