大鼠肢体缺血再灌注对脊髓神经元细胞凋亡的影响及机制探讨 被引量:5

摘要 目的:观察在大鼠肢体缺血再灌注脊髓神经元损伤的过程中,细胞凋亡以及Bcl-2、Caspase-3蛋白质表达的变化。方法:复制实验性大鼠肢体缺血再灌注损伤模型,观察脊髓L1-5节段的病理学改变,TUNEL法检测细胞凋亡情况,免疫组化检测脊髓神经元Bcl-2、Caspase-3蛋白质表达的变化。结果:肢体缺血再灌注后Bcl-2、Caspase-3表达明显增多,Bcl-2在12h达高峰,之后下降;Caspase-3在24h达高峰,24h的凋亡细胞数最多,Caspase-3的表达与凋亡细胞数呈正相关关系,r=0.946。结论:肢体缺血再灌注后出现脊髓神经元损伤,并有细胞凋亡发生。 Objective:To observe the expressive changes of the cell apoptosis, Bcl-2 and Caspase-3 during the process of spinal cord neuron injury of limbs isehemia-reperfusion (LIR) in rats. Methods:The limbs ischemia-reperfusion injury model of experimental rats was duplicated to observe the pathologic changes of the segment L1-5 of the spinal cord by HE staining, to detect the cell apoptosis by TUNEL method and to determine spinal cord neurons Bcl-2 and Caspase-3 protein by immunohistochemistry. Results:The expression of Bcl-2 and Caspase-3 followed LIR was significantly increased. Bcl-2 got the peak value at 12h and then was gradually decreased. Caspase-3 got the peak value at 24h and the numbers of cell apoptosis were the most at 24h. The expression of Caspase-3 and the numbers of cell apoptosis were positive correlation and r = 0.946. Conclusion: Spinal cord neuron injury may appear after limbs ischemia-reperfusion(LIR) and the cell apoptosis may occur.
