摘要
应用基因工程方法将金属硫因(MT)表达到B72.3单抗的Fc段上,制备出双功能杂交分子B72.3MT-1。采用99mTc直接标记B72.3MT-1显示标记过程简便、快速、标记率高,标记物稳定性好,不易脱落,适于药盒化生产。体外细胞结合实验、动物体内分布实验和病人RI研究证明,99mTc-B72.3MT-1与99mTc-B72.3两者所得结果基本一致,均可同样用于肺腺癌和胃腺癌的定位诊断。
おo evaluate the clinical diagnostic value of 99m Tc labeled bifunctional hybrid protein B723 MT1.The bifunctional hybrid protein B72.3 MT1 was produced by the fusion of mouse metallothionein1 cDNA gene into CH2 region of B72.3 heavy chain cDNA fragment using the recombinant DNA technology.The 99m Tc direct labeling method to B72.3 MT1 in cell binding assay in vitro,animal biodistribution and patient RⅡ were in accord with these of 99m TcB72.3 RⅡ of 99m TcB72.3 MT1 may be applied as a valuable method in the diagnosis of lung adenocarcinoma,gastroadenocarcinoma and may be useful for differentiating adenocarcinoma from nonadenocarcinoma.
