摘要
目的:原代培养大鼠耳蜗血管纹血管内皮细胞,为血迷路屏障研究提供体外细胞模型。方法:显微解剖分离新生Wistar大鼠耳蜗血管纹组织,运用胰蛋白酶消化与机械分离组织块相结合的方法培养细胞,免疫组织化学染色鉴别细胞来源。结果:部分血管纹组织块培养48h后,自边缘生长出零散的细胞,逐渐增殖并形成致密融合的单层多边形细胞的细胞集落,经传代纯化的细胞具有内皮细胞培养典型的"铺路石样"外观,免疫组织化学检测内皮细胞标志性抗原Ⅷ因子,细胞胞浆中呈棕黄色阳性反应。结论:本实验培养出大鼠耳蜗血管纹血管内皮细胞,为血迷路屏障体外研究提供了细胞模型。
Objective:To provide an invitro cell model for blood-labyrinth barrier by primary culturing cochlear capillary endothelial cells from stria vascularis of rats. Methods:Cochlear stria vascularis were acquired from neonatal Wistar rats with micrergy,the cells were cultured by trypsin digestion with tissue nubbles cultivation,then the immunocytochemistry was employed to classify cell types.Result:48 hours after tissue nubbles cultivation of stria vascularis,a few cells were seen disseminately around partial tissue nubbles,and proliferated to massive colonies composed of more cells.These cells were mainly polygen,monolayer confluent. After passage and purification,the cells showed typical "flagstone" appearance as cultured endothelial cells.According to immunocytochemiscal dictection of facter Ⅷ ,which is marked antigen of endothelial cell,we observed that the cultured cells show positive reaction with cytoplasmic brown pigment.Conclusion:The method used in this study can obtain cochlear capillary endothelial cells from the stria vascularis,which can be used as an in vitro model for blood-labyrinth barrier.
