摘要
目的建立HPLC-ELSD法测定罗红霉素的含量。方法采用Phenomenex C18色谱柱,以甲醇-0.2%三氟醋酸(TFA)溶液(50:50)(用三乙胺调节pH至6.5)为流动相,柱温为室温,流速为1.0ml/min,以蒸发光散射检测器检测,漂移管温度:90℃,空气作为载气,流速为1.6L/min。结果罗红霉素在2.004~10.02μg范围内与相应峰面积呈良好线性关系(r=0.9996,n=5),平均回收率为99.11%,RSD为0.5%(n=5)。结论方法准确,专属性强,该法可做为罗红霉素含量测定的有效方法。
Objective An HPLC method for the content determination of roxithromycin was established. Methods The determination was performed on Phenomenex C18 column with the mobile phase consisted of methanol -0.2%trifluoroacetie acid (TFA) solution (50: 50)(pH was adjusted to 6.5 with triethylamine) at room temperature. ELSD was used as detector, the drift tube temperature was 90℃ and eluant gas was air with the flow rate of 1.6L/min. Results The method showed a good linear relationship within the range of 2.004 - 10. 02μg ( r = 0. 9996, n = 5). The mean recovery was 99. 11% with RSD = 0. 5% (n = 5). Conclusion The method is simple, accurate and could be used to content determination of roxithromycin.
