独角莲抑制乳腺癌MCF-7细胞增殖和诱导凋亡的作用研究 被引量:15

摘要 为探讨独角莲对人乳腺癌MCF-7细胞增殖和凋亡的作用,将不同浓度的独角莲根茎水提物(The aqueous extract from dried powdered rhizomes of Typhonium giganteum Engl.,AEoTGE)处理MCF-7细胞24,48,72h后,应用四甲基偶唑蓝(Methyl thiazolyl tetrazolium,MTT)比色试验、倒置光学显微镜观察、流式细胞术(Flow cytometry,FCM)及琼脂糖凝胶电泳法检测细胞增殖和凋亡情况。结果表明:AEoTGE能够显著抑制MCF-7细胞增殖,且在一定的浓度范围内呈时间和浓度依赖性,并可诱导其细胞凋亡。30g·L-1AEoTGE处理24,48,72h后,MCF-7细胞凋亡率分别为8.34%、12.1%和12.6%,其凋亡程度与时间呈正相关,S期(DNA合成期)的细胞在处理72h后分布显著增加。可见,独角莲对MCF-7细胞有抑制增殖和诱导凋亡作用,其抗肿瘤机制与诱导凋亡有关。 The effects of the aqueous extract from dried powdered rhizomes of Typhonium gigantettm Engl.,(AEoTGE) on proliferation and apoptosis of MCF-7 cells were studied. MCF-7 were incubated with AEoTGE at different concentrations for 24h, 48h and 72h, respectively.The effects of AEoTGE on cell proliferation were measured with Methyl thiazolyl tetrazolium (MTY) colormetric assay and cell apoptosis with inverted optical microscope, flow cytometry (FCM)and agarose gel electrophoresis. AEoTGE inhibited the proliferation of MCF-7 cells in a time and dose dependent manner. The apoptotic rate of ceils treated with 30 g·L^-1 AEoTGE were 8.34%,12.1% and 12.6% in 24h, 48h and 72h intervals. Cells of MCF-7 was apparently in S phases by AEoTGE in time depending manner. Typhonium giganteum Engl could inhibit proliferation and induce apoptosis in MCF-7 and might play a role in treatment of breast cancer.
