摘要
用液态发酵法让黑曲霉产果胶酯酶,酶活力为467U/ml,果胶酯酶的最佳pH为4.0,温度50℃。用自制果胶酯酶处理含DCS水,CD值最多降低30%,并使得DCS水粒径分布改变,浊度降低,电导率增大。最适加入量为150U/100ml,继续增大酶用量会导致CD值回升。最佳时间为60min,继续延长处理时间,CD值、浊度和电导率变化不大。
Through liquid fermentation method, pectinesterase was produced from aspergillus niger with the enzymic activity of 467U/ml. The optimal temperature of pectinesterase was 50℃ and optimal pH was 4.0. Treating DCS water with pectinesterase made in lab can decrease CD value by 30% at most, change particle size distribution, reduce turbidity and increase conductivity of DCS water. The most appropriate addition of pectinesterase was 150U/100ml and more pectinesterase added resulted in the return of CD value. The best treating time was 6.0min and time prolonging resulted in no obvious change of CD value, turbidity and conductivity.
