摘要
目的研究大鼠脑缺血再灌注(IR)脑内巢蛋白(Nestin)和胶原纤维酸性蛋白(GFAP)表达变化及骨髓间充质干细胞(MSCs)移植对它们的影响。方法采用微动脉夹夹闭左侧颈总动脉的方法制作大鼠脑缺血再灌注模型;MSCs治疗组于缺血再灌注后1天、4天及8天经尾静脉注射MSCs(1×106)后分别在缺血再灌注3天、7天、14天的各时间点处死动物,取前脑组织,用免疫组织化学方法检测各组大鼠脑内Nestin和GFAP的表达情况;用SPSS统计分析。结果免疫组化检测MSCs干预组在缺血10min后再灌注3d、7d、14d时,大鼠脑组织中Nestin和GFAP的表达增强,相同时间点比较有显著性差异(P<0.05);结论MSCs可激活大量的星形胶质细胞,保护神经元,促进其修复,有利于功能的恢复。
Objective To study the expressions of Nestin and GFAP protein and the effects of transplantation of mesenchymal stem cells(MSCs) on the expressions of Nestin and GFAP protein in brain tissue following transient cerebral isehemic reperfusion in rats. Methods A cerebral ischemia-reperfusion model in rats was estabished by clamping left common carotids artery. The rats in treated group were injected 1 × 10^6 MSCs via tail vein individually at the 1st day,4th day and 8th day. The expressions of Nestion and GFAP protein in brain tissue were detected by using immunohistoehemistry technique at the 3rd day,7th day and 14th day after ischemic reperfusion(IR). Experimental results were comparatively analyzed with statistic soft package of Spssl 1.0. Results The results revealed that the expression of Nestin and GFAP protein in neurons and gliacytes of rats in MSCs treated group was evidently increased at the 3rd day,7 th day and 14 th day ( P 〈 0.05 ). Conclusions Expressions of Nestin and GFAP protein in neurons and gliacytes of gerbils were evidently increased. This suggests that MSCs may activate astrocytes for the neuron protection and promote repairing of neuron.
