内脏脂肪对发育期大鼠骨密度的影响及其机制探讨

摘要 目的:了解高脂喂养对于发育期大鼠骨形成的影响,并探讨骨密度(BMD)与内脏脂肪及血清瘦素间的相关性。方法:两组5周龄雄性Wistar大鼠,分别给予普通饲料(CON组)和高脂饲料(HF组)喂养12周,双能X线仪(DXA)检测BMD,ELISA法检测血瘦素值。结果:两组大鼠体重无明显差异,但HF组内脏脂肪/体重比明显增高;HF组股骨BMD明显低于CON组,而血清瘦素水平高于CON组,内脏脂肪比及瘦素与股骨BMD呈负相关。结论:给予发育期大鼠高脂喂养会导致腰椎骨密度下降,其中的机制可能与内脏脂肪的堆积有关,同时还受脂源性瘦素的调节。 To investigate the influence of high fat (HF) diet on the bone mineral density of growing rats, and to explore the correlation among visceral fat, serum leptin and bone mineral density. Methods: Twelve 5-week-old male Wistar rats were fed with high fat diet for 12 weeks. Bone parameters were assessed by dural energy X ray assay (DXA), and serum leptin was detec ted by ELISA. Results: HF rats had the same body mass as contols, but the visceral fat/body mass ratio was higher, and bone mineral density (BMD) was lower. Significant correlations were noted between visceral fat, leptin and BMD in rats. Conclusion: High fat diet can damage bone metabolism in growing rats, and has a relationship with accumulation of visceral fat and the change of serum leptin.
