摘要
改进了对硫酸氨基葡萄糖检测的反相高效液相色谱方法.柱前用异硫氰酸苯酯(GITC)衍生,在254nm紫外波长处紫外检测洗脱液.色谱柱使用C18柱,流动相是由MeOH/H2O/CH2COOH(体积比为20:79.96:0.04)组成,流速为0.6mL/min.结果显示,在氨基葡萄糖含量为8~100μg/mL范围内呈线性关系(r=0.9985),样品平均回收率为98.72%,RRSD=0.473%(n=5).该方法简便、准确,可用于氨基葡萄糖硫酸盐的检测.
The RP-HPLC method for the determination of glusamine sulfate was improved. The phenylisothiocyanate (GITC) was used as a precolumn derivatizing reagent in RP-HPLC with UV detection at 254 nm. The derivatized solution was quantified on C18 column using V(MeOH) : V (H20) : V (CH3COOH) = 20 : 79.96 : 0.04 as mobile phase at a flow rate of 0.6 mL/min. The linear range of standard curve was 8-100μg/mL ( correlation coefficient r = 0. 998 5 ). The average recovery of the sample was 98.72%, and the relative standard deviation ( RSD ) was 0. 473G (n = 5 ) . The method is simple, reliable and can be used for determination glusamine sulfate.
