不同来源珍珠粉中氨基酸类成分的HPLC-ESI-MS/MS分析与评价 被引量:2

Analysis and Evaluation of Amino Acids in Margarita Powders from Different Habitats by HPLC-ESI-MS/MS

摘要 目的:建立HPLC-MS/MS法同时测定珍珠粉中14个氨基酸类成分含量,评价不同来源珍珠粉的质量。方法:采用TSKgel Amide-80(15 mm×2 mm,3μm)分析柱,以0.2%甲酸溶液-乙腈为流动相,梯度洗脱;流速为0.2 mL/min;检测器为QTRAP 5500质谱仪;对50批不同来源珍珠粉中14个氨基酸类成分进行检测。结果:14个氨基酸类成分在一定的浓度范围内与峰面积的线性关系良好,检测限为0.0010~0.0228μg/mL,日内精密度、日间精密度、重复性、稳定性试验结果的RSD依次为1.23%~3.77%、1.86%~3.06%、1.53%~2.82%、0.78%~2.54%,平均加样回收率为82.00%~112.70%。浙江诸暨、太湖、湖北汉阳的珍珠粉氨基酸总含量较高,而浙江义乌收集的珍珠粉的氨基酸总含量较低,均<1.0 mg/g。结论:该方法操作简便、准确,可用于珍珠粉中氨基酸类成分的测定及质量评价。 Objective:To establish HPLC-MS/MS method for simultaneous content determination of fourteen amino acids in Margarita powders,to evaluate the quality of Margarita powders from different habitats.Methods:The fourteen amino acids was separated with a TSKgel Amide-80(15 mm×2 mm,3μm)column,the mobile phases was 0.2%formic acid solution-acetonitrile with gradient elution,the flow rate was 0.2 mL/min.The detector was QTRAP 5500 mass spectrometer.Fourteen amino acids in 50 batches of Margarita powders from different habitats were detected.Results:The linear relationships between the contents of fourteen amino acids and the peak areas were good in a certain range,and the detection limit was 0.0010~0.0228μg/mL.The RSDs of intra-day precision,inter-day precision,reproducibility and stability tests were 1.23%~3.77%,1.86%~3.06%,1.53%~2.82%,0.78%~2.54%,respectively.The average recoveries were 82.00%~112.70%.The total content of fourteen amino acids of Margarita powders from Zhejiang Zhuji,Taihu and Hubei Hanyang was higher,while the total content of amino acids of Margarita powders collected from Zhejiang Yiwu was lower,which was less than 1.0 mg/g.Conclusion:The method is simple and accurate,and can be used for the determination and quality evaluation of amino acids in Margarita powders.
