摘要
目的观察雷公藤甲素对子宫内膜癌细胞株HHUA增殖的影响。方法分别采用MTT法及流式细胞仪观察雷公藤甲素对子宫内膜癌细胞株HHUA增殖、细胞周期以及凋亡的影响。结果MTF实验结果显示,5~160ng/mL的雷公藤甲素均能抑制HHUA细胞的生长且生长抑制作用呈剂量及时间依赖性;流式细胞仪检测显示,雷公藤甲素(40ng/mL、80ng/mL)处理HHUA细胞48h后,能诱导HHUA细胞发生细胞周期阻滞,阻滞部位在S期,并诱导其发生凋亡。结论雷公藤甲素能抑制人子宫内膜癌细胞株HHUA细胞的生长,将其阻滞在S期,并诱导其发生凋亡。
Objective To examine the effect of triptolide on the growth of human endometrial cancer cell line. Methods MTT experiments were performed to examine the inhibiting effect of triptolide on the proliferation of HHUA cells, and flow cytometry was used to analyze cell cycles and apoptosis peak. Results The resuits of MTT experiment showed that triptolide inhibited proliferation of HHUA cells in a concentration and time- dependent manner (5 -160 ng/mL). Flow cytometry showed that 48 hours after using triptolide to treat (40 and 80 ng/mL) HHUA cells, cell cycle was blocked at S phase, and an obvious apoptosis peak appeared before G1 stage. Conclusion Triptolide can inhibit the proliferation of HHUA cells, block HHUA cells at S phase and induce apoptosis of HHUA cells.
