摘要
应用MTT法测定一叶秋碱对人早幼粒细胞白血病细胞HL60、人红白血病细胞K562、人骨肉瘤细胞HOS-8603、人脑胶质瘤细胞SHG-44的增殖抑制作用;体内采用小鼠移植性肿瘤模型,观察一叶秋碱对艾氏癌实体型(EC)抑癌作用。结果:一叶秋碱对4株肿瘤细胞的增殖均有抑制作用,药物作用24h的IC50分别为6.18、26.8、68.4、120.9μg/ml;对EC平均抑瘤率为25.3%~62.2%,与环磷酰胺合用仰瘤作用相加。
To studay the anti - tumor effect of Securinine (SEC ) in vivo and in vitro. METHOD: In order to investigate the anti - tumor effect in vivo , SEC was administrated singlely or in combination with CTX to treat transplated tumor EC. The changes of tumor weight and average weights of mice were observed. Using MTT assay: the inhibitory effect of SEC on human leukemia cell line HL60 ana K562, human brain glioma cell line SHG - 44 and human osteosarcoma cell line Hos-8603 were determined. RESULT: SEC siginificantly inhibited the growth of EC in vivo. The average inhibitory rate of SEC was 25 .3 % - 62. 2%. In combibiantion with CTX, the effect was much more marked than in independent adminstration that was. The proliferation of four human tumor cell line yas inhibited after incubation with SEC for 24h. The values of IC50 were 6. 18μg/ml, 26. 8μg/ml,68 .4μg/ml and 120 .9μg/ml respectively. CONCLUSION: SEC has a certaion antitumor effect.
