摘要
在粳稻花药培养过程中,花粉植株的诱导频率受接种材料的基因型、花粉发育时期和培养基成分等因素的影响.杂交种的诱导率高于纯合品种,接种最佳时期为花粉发育单核中晚期,诱导培养基以N6+2.4—D2mg/l+KT0.5mg/l+蔗糖5%+琼脂粉0.7%;分化培养基以Ms+BA2mg/l+NAA0.2—0.5mg/l+蔗糖3%+琼脂粉0.7%;壮苗培养基以1/2MS+微量+铁盐+NAA0.1—0.2mg/l+多效唑2mg/l+蔗糖2%+琼脂0.
In the anther culture of Japonica Rice,the induction rate of pollen plantlets was affected by the genotypes of materials, developmental stage of anthers,formulae of culture medium and so on. Hybrids had a higher induction rate as compared with homozygous varieties.The suitable developmental stage of the microspores in the anthers was in the mid-or-late-uninucleate.Better medium formulae were N6+2mg/l2.4-D+0.5mg/l KT+5% sucrose+0.7% agar for dedifferentiation and Ms+2mg/lBA+0.2-0.5mg/l NAA+3% sucrose+0.7%agar for differentiation.To make the plantlets strong,you should transfer them onto 1/2 Ms (allFe-salt)+0.1-0.2mg/l NAA+2mg/l MET+2% sucrose+0.7% agar.
