棉枯、黄萎病抗性的PPO同工酶标记鉴定

摘要 利用聚丙烯酰胺凝胶电泳技术测试分析了棉花萌动种子及棉株感病前后叶片的多酚氧化酶(PPO)同工酶。结果表明,罹病反应中叶片PPO同工酶谱的变化差异可作为鉴定抗枯、黄萎病大小的生化指标;棉花萌动种子PPO同工酶在不同抗枯黄萎病品种间及品种内个体之间表达均存在多态性,确认了分别与抗枯萎病和抗黄萎病性状相关的两条标记酶带。根据这两条标记酶带频率的大小,可鉴定棉花品种(系)的抗性水平。同工酶测试分析后的种子经室内培养后在田间仍能正常生长,其标记酶带与田间抗病性表现一致,因而可作为筛选抗病品种的一种室内方法。 Polyphenol oxidase (PPO)isozymes of germinative cotton and leaves were invesigated by polyacrylamide gel electrophoresis (PAGE) The results showed that the change of PPO isozymes in leaf can be used as biochemical marker in identifing the resistance of cotton to Fusarium and Verticillium after the leaf displayed obvious wilt reaction,polymorphism of PPO isozymes in germinative cotton seeds exists not only in different varieties,but also in individuals of the identical variety Two isozyme bands relating to resistance of cotton varieties to Fusarium and Verticillium were respectively confirmed,and according to the frequency of two isozyme bands,the resistance of cotton to Fusarium and Verticillium can be appraised The seeds tested by PAGE can still grow and develop in field normally
