摘要
目的研究神经生长因子(nerve growth factor,NGF)和前列腺素E1(prostaglandin E_(1))对周围神经损伤后运动功能恢复的促进作用。方法清洁级SD大鼠,制备坐骨神经夹毁模型,术后每日分别局部给予一定剂量的NGF、PGE_(1)和等渗盐水共14天。术后20天制备再生坐骨神经-胫前肌在体标本,用三导记录仪记录肌肉收缩张力曲线(CTCM),术后每日或隔日测夹毁侧和正常侧后足足印长度(PL)和足趾伸张距离(TSD),以夹毁侧的PL值和TSD值接近正常侧PL值和TSD值作为该项运动功能恢复正常的标志。结果NGF和PGE_(1)均有效地增进肌肉收缩张力,加快肌肉收缩峰值速度和减轻肌肉萎缩率,明显缩短PL和TSD恢复正常的时间。结论外源性NGF能明显促进周围神经损伤后运动功能恢复。
Aim To study the effect of the nerve growth factor(NGF)and prostaglandin E_(1)(PGE_(1))on the motor function recovery of the sciatic nerve following crush injury.Methods SD rats with the sciatic nerves crushed were randomized into four groups:Sham operation control,crush control,crush PGE,and crush NGF group,and the animals in the lateral three groups were locally injected sterile saline group,PGE,and NGF once a day after axotomy for 14 days,respectively.The motor function recovery was indicated by the recovery of the toe spreading reflex and the footprint length and the curve of the muscular constractile tension(CTCM).Results Both NGF and PGE_(1)could significantly shorten the recovery time of toe spreading reflex and the footprint length,promote the muscular constractile strength and velosity,and decrease the percentage of muscular atrophy.However,the effect of PGE_(1)on motor function recovery was lower than that of NGF.Conclusion Exogenous NGF can significantly enhance the motor function recovery of the sciatic nerve following axotomy in the rats,and PGE_(1)can also partially mimic the role of NGF to recover the sciatic nerve injury.
