瑞香狼毒药效组分抑制小鼠皮下移植瘤H22的实验研究 被引量:10

Antitumor activities of active components alignments isolated from Stellera Chamaejasme L. in H22-bearing mice

摘要 目的:考察两种体内给药途径瑞香狼毒药效组分对小鼠肝癌移植瘤H22抑制作用强度与量效关系及对荷瘤小鼠免疫器官的影响。方法:建立小鼠皮下肝癌移植瘤H22实验动物模型,采用灌胃与腹腔注射两种给药途径,瑞香狼毒药效组分Zp1102、Zp1103、Zp1105、Zp1111、Zp1113在小于1/3-1/5LD50的剂量范围内,观测各受试药对移植瘤H22的抑制作用及其量效情况,并测算实验小鼠胸腺与脾脏指数。结果:瑞香狼毒各药效组分对小鼠皮下移植瘤H22具有较强的抑制作用,并呈一定剂量依赖关系,其中Zp1102、Zp1103经灌胃给药2g/kg(折合成生药量)时能达到35%左右的抑瘤率;Zp1105灌胃给药效果较差(16g/kg时仅能达到35%左右的抑瘤率),而腹腔注射给药法250mg/kg时就可以达到38%的抑瘤率;Zp1111在250mg/kg剂量时对皮下移植瘤H22的抑瘤率达48.35%。多数药效组分对荷瘤小鼠的体质量与免疫器官系数较溶剂对照组有所下降,但其影响比阳性对照药要小。结论:瑞香狼毒药效组分对小鼠皮下移植瘤H22具有较强的抑制作用,且用药安全性较好,其中Zp1111抑瘤作用最强;腹腔注射给药在较低剂量即可显示良好的抑瘤作用。 Objective: To investigate the antitumor activity of active components alignments isolated from Stellera chamaejasme L. in vivo and their impacts on immune organs of the H22-bearing mice. Methods: To study the antitumor activities of the active components alignments which named Zp1102, Zp1103, Zp1105, Zp1111, Zp1113 on H22-bearing mice judged by the inhibitory rate of tumor at the dosage of 1/3-1/5 sub-LD50 (Median fatal dose), and to survey their toxicity by calculating the index of thymus-gland/spleen wet weight and the loss in weight of mice. Results: The active components alignments exhibited significant antitumor activities in vivo and showed obvious dose-dependent response, in which the Zp1111 was most active, with the tumor inhibitory rate 48.35% at a low dosage by peritoneal injection. Some active components alignments impacted the immune organs compared with the control groups but majority of them more slightly than that of the positive control ones. Conclusion: The active components alignments isolated from Stellera chamaejasme L. showed effective antitumor activation in vivo, in which the Zp1111 was most active,and the route of administration by peritoneal injection displayed stronger effects with a lower dosage.
