同型半胱氨酸对内皮祖细胞增殖 黏附和凋亡的影响

摘要 目的观察不同浓度同型半胱氨酸(Hcy)对内皮祖细胞(EPCs)功能的影响。方法密度梯度离心法获取单个核细胞,培养7d后,收集贴壁细胞。在多波长激光共聚焦显微镜下,呈异硫氰酸荧光素标记的荆豆凝集素(FITC-UEA-I)和DiI标记的乙酰化低密度脂蛋白(DiI-acLDL)双染色阳性者为正在分化的EPCs,流式细胞仪检测其表面标志进一步鉴定EPCs。不同浓度同型半胱氨酸组(10、50、100、200μmol/L)干预24h,分别测定EPCs的增殖、黏附和凋亡。结果与对照组比较,Hcy组呈剂量依赖性地损害EPCs的增殖、黏附,促进其凋亡。200μmol/L的Hcy组对EPCs影响最为显著(P<0.01)。结论Hcy损害EPCs的功能。 Objective To investigate the effects of homocysteine on proliferation, adhesion and apoptosis of endothelial progenitor cells (EPCs). Methods Density gradient centrifugation was used to obtain mononuclear cells. After culture for 7 days, the adherent cells were collected. Under multi-wavelength laser scanning microscope, cells stained positive for both FITC-UEA-I and DiI-acLDL were identified as differentiating EPCs. Flow cytometry were used to further identify the surface markers on EPCs. The EPCs were determined for their proliferation, adhesion and apoptosis after incubation with different concentrations of homoeysteine (10, 50, 100, 200 μmol/L) for 24 h. Results Compared with control group, the Hey-treated EPCs showed impaired proliferation and adhesion but enhanced apoptosis in a dose-depending manner. Effects of 200 μmol/L Hcy appeared the most significant on EPCs (P〈0.01). Conclusion Hey may have harmful effects on EPC function.
