摘要
目的研究黄芪对人外周血内皮祖细胞(EPCs)增殖、细胞周期的影响。方法采用密度梯度离心法获取外周血单个核细胞,培养7天后,收集贴壁细胞并加入不同浓度黄芪注射液(0.2、2、20和200mg/ml)干预一定时间(6、12、24和48小时)。激光共聚焦显微镜鉴定人外周血EPCs,噻唑蓝比色试验(MTT法)检测黄芪对人外周血EPCs体外增殖作用,流式细胞仪检测细胞周期的变化。结果黄芪明显促进人外周血EPCs的增殖,呈一定的量效时效关系;流式细胞仪分析表明,黄芪作用后,细胞周期中S期、G2/M期细胞比例增加,G0/G1期细胞相对比例减少(P<0.01)。结论黄芪对人外周血EPCs增殖有显著促进作用,其作用机制可能是通过促进DNA合成,增加进入S期细胞数目来实现的。
Objective To study the effect of Huangqi (Radix Astragali seu Hedysari) on the proliferation and cell cycle of endothelial progenitor cells (EPCs) of human peripheral blood. Methods The mononuclear cells were isolated from human peripheral blood by density gradient eentrifugation. After 7 days' culture, the anchorage-dependent cells were collected and the Huangqi Injectio (0.2, 2, 20 and 200mg/ml) was injected for intervention (for 6, 12, 24 and 48h), The laser scanning eonfoeal microscope was used to determine the EPCs of human peripheral blood, the methyl thiazolyl tetrazoliym (MTT) assay was applied to test the proliferation of EPCs in vitro, and the flow cytometry (FCM) was adopted to detect the changes of cell cycle. Results Huangqi promoted the proliferation of EPCs remarkably, showing a time- and dose-effect relation. The FCM analysis showed that the S-phase and G2/M-phase rate increased but G0/G1-phase rate decreased relatively (P〈0.01). Conclusion Huangqi functions to promote the proliferation of EPCs remarkably and its mechanism may be promoting DNA synthesis and increasing the number of cells entering S-phase.
