摘要
目的观察天花粉蛋白经由TCR/CD3介导的信号传递途径抑制T细胞活化增殖的效应。方法用各种不同的剌激信号来激活外周血淋巴细胞,然后观察天花粉蛋白对其增殖抑制的影响。接着测定天花粉蛋白对CD3单抗剌激后T淋巴细胞内一些信号传导物质变化的影响。结果天花粉蛋白抑制经由TCR/CD3信号传导途径诱发的淋巴细胞增殖。进而发现细胞内游离钙离子的升高,蛋白激酶C的转位和细胞内蛋白酪氨酸磷酸化受天花粉蛋白的抑制。结论天花粉蛋白可通过TCR/CD3的信号传导途径抑制T细胞增殖。
Trichosanthin can inhibit the lymphoproliferation in vitro via immunosuppression. In this paper,PBMC were stimulated with different stimuli and the mechanism of inhibition was studied.Our results showed that the inhibition of lymphoproliferation by Trichosanthin was resulted from interfering with the signaling pathway directed by TCR/CD3 complex.Moreover, there were alternations of signaling events in antiCD3 stimulated PBMC after Trichosanthin treatment,which included a raising of cytosolic Ca2+,the suppression of PKC translocation and a decreased phosphorylation of protein tyrosine kinase.It suggests that Trichosanthin can suppress T cell activation by means of interfering with signal transduction.
