摘要
利用原生质体融合技术进行耐温的克鲁维酵母(KluyveromycesY034)与酿酒酵母(SaccharomycescerevisiaeA001)的属间融合,获得属间融合子AY068。该融合株在45°C下进行高温乙醇发酵,结果表明融合株表达了双亲耐温和产酒率高的特性。通过正交试验优化培养基成分,表明蛋白胨用量对菌体热抗性差异显著。比较了不同温度条件下,菌体生物量、糖利用率、pH因素与酒精产率的相互关系,为菌株工业生产提供了理论依据。
Fusant AY068 was obtained by protoplast fusion between Saccharomyces cerevisiae A001 and Kluyveromyces Y034, and it expressed excellent properties of parent strains such as thermotolerance and high ethanol yeild at 45℃. The result of orthogonal design showed that peptone played an important role in thermotolerance of strain. Meanwhile, several kinds of factors such as cell increase, ethanol yeild, glucose utilization. pH and so on were researched at different fermentation temperature.
