摘要
[目的]初步研究甘露聚糖酶基因3′端对甘露聚糖酶功能的重要性。[方法]以甘露聚糖酶的二级结构为基础,用PCR扩增的方法获得3′端特异性缺失的甘露聚糖酶DNA片段,连接到载体上,表达并鉴定酶的活性。[结果]删除甘露聚糖酶基因3′端246 bp后甘露聚糖酶依然具有酶活性。[结论]甘露聚糖酶基因3′端的部分序列不是酶具有功能所必需的。
[ Objective]The paper aimed to study the importance of 3' sequence for the function of β-mannanase. [ Method]β-mannanase DNA segement lack of 3' sequence was obtained by PCR amplification according to its secondary structure. The DNA was linked to vector for expressing and identifying the activity of β-mannanase. [ Result ] After 246 bp was shortened from the 3'sequence, the activity of β-mannanase was still available. [ Conclusion] Part of the 3' sequence was not inevitable for the function of β-mannanase.
