摘要
目的探讨血小板膜糖蛋白(GP)Ia基因C807T的多态性与患者发生阿司匹林抵抗的相关性.方法330例心脑血管病患者服用阿司匹林(100mg/d)至少7d以上,用二磷酸腺苷(adenosinediphosphate,ADP)和花生四烯酸(araehidonicacid,AA)作为诱导剂测定血小板聚集功能,找出阿司匹林抵抗(aspirinresistance,AR)患者,用PCR法确定血小板膜糖蛋白Ⅰa基因C807T的基因型.结果等位基因纯合子807(T/T)型和等位基因T在阿司匹林半抵抗与敏感人群中分布比较差并有统计学意义(P〈0.05).结论血小板膜糖蛋白Ⅰa/Ⅱa的纯合子等位基因807T/T型与阿司匹林抵抗的发生相关,阿司匹林抵抗可能与遗传因素有关.
Objective To investigate the associativity between platelet membrane glycoprotein Ⅰa/Ⅱa polymorphism C807T and aspirin resistance. Methods 330 stable cardiovascular and cerebrovascular diseases patients took aspirin(100mg/d for ≥7 days ). Detected platelet aggregation function using adenosine diphosphate (ADP) and arachidonic acid (AA). Genotyping of platelet membrane glycoprotein Ⅰa/Ⅱa C807T polymorphism was formed using a polymerase chain reaction analysis. Results The genotype 807(T/T) of aspirin semiresponders (ASR) were significantly different from aspirin sensitive(AS) patients (P〈0. 05). Conclusion Platelet membrane glycoprotein Ⅰa/Ⅱa homozygote 807T/T is related to aspirin resistance. Aspirin resistance would be influenced by genetics.
