摘要
建立一种应用异硫氰酸苯酯将,L-瓜氨酸和L-鸟氨酸柱前衍生后进行定量分析的高效液相色谱法.实验结果表明,瓜氨酸在0.2~2.0mg/mL范围内线性回归显著(R^2=0.9998),平均回收率99.87%;鸟氨酸在0.2-2.0mg/mL范围内线性回归显著(R^2=0.9997),平均回收率100.92%.应用该方法对发酵液中L-瓜氨酸和L-鸟氨酸含量进行测定取得了满意的结果,能有效指导谷氨酸棒杆菌L-瓜氨酸的发酵生产.
A method for reversed-phase high performance liquid chromatography (HPLC) with phenylisothio cyanate precolumn derivation was developed for the determination of L-citrulline and L-omithine. The calibration curves of L-citrulline and L-omithine were linear over the range of 0. 2 - 2. 0 mg/mL, and the recovey rates were 99. 87% and 100.92% (n =6), respectively. It can be used as one of the direct and reliable means for quantitative determination of L-citrulline and L-ornithine. Thus, it plays an instructive role in the production of L-citrulline by corynebacterium glutamicum.
