RP-HPLC法测定青果鲜果肉中的没食子酸含量 被引量:7

摘要 在色谱柱为NucleosilC18反相柱(250mm×4mm),流动相A为体积分数20%的甲醇,流动相B为体积分数30%的乙腈,A和B流动相用体积分数50%的磷酸调pH至3.2±0.05,流动相C为乙腈,三元梯度洗脱,流速0.8~1mL/min,柱温35℃,分析波长272nm条件下,应用反相高效液相色谱法测定了广东产15个品种青果鲜果肉中的没食子酸含量.结果表明,没食子酸线性范围为0.3906—25.0000μg/mL,标准曲线y=62.662x-16.805,相关系数殿=0.9996.标准品保留时间和峰面积的相对标准偏差分别为1.7614%和2.7736%.标准品回收率平均数为99.09%(n=5),回收率相对标准偏差为3.298%.15个品种青果果肉中均含有没食子酸,含量在57.9584~230.7373μg/g鲜质量之间,以土纳甜种品种的含量最高,香种含量最少,相对标准偏差为0.060%~1.938%. A reverse-phase high-performance liquid chromatography (RP-HPLC) method was used for measuring gallic acid content in flesh of 15 Chinese olives varieties. Samples were injected onto reversedphase Nucleosil C18 column (250 mm ×4 mm i. d. ). Mobile phase A and B were 20% methanol and 30% aeetonitrile respectively. Both of them were adjusted to pH3.2 with 50% phosphoric acid. Acetoni- trile was set as mobile C. Triple gradient elution proceeded at the flow rate 0. 8-1 mI./min. Operation conditions were as follows: column temperature 35 ℃, detection at 280 nm of wavelength. The results showed that linear calibration of gallic acid was within 0. 390 6-25.000 0 μg/mL with correlation (r) of 0. 999 6. Regression equation was acquired as y =62. 662x-16. 805. Relative standard deviation (RSD) of retained time and peak area of standard sample were respectively 1. 761 4% and 2. 773 6%. Average recovery of standard sample was 99.09% (n =5) with RSD of 3.298%. Gallic acid was detected in all olive samples with the content within 57.958 4 -230. 737 3 μg/g fresh mass (FM). According to the content of gallic acid, the richest variety was Tunatianzhong while the least was Xiangzhong with RSD of 0. 060%-1. 938% .
