PTEN和诱导型一氧化氮合酶蛋白在子宫内膜癌中的表达 被引量:1

摘要 目的研究子宫内膜癌组织中PTEN和诱导型一氧化氮合酶(iNOS)蛋白的表达,并探讨其临床意义以及二者的相关性。方法采用免疫组织化学EnV isionTM法,检测55例子宫内膜癌、24例子宫内膜不典型增生和20例正常子宫内膜石蜡切片中PTEN和iNOS的表达。结果与正常子宫内膜(阳性表达率100%)比较,PTEN在子宫内膜不典型增生及子宫内膜癌组织中阳性表达率逐渐降低,分别为91.67%和60%(P<0.05);iNOS阳性表达率依次为5.00%、8.33%和76.36%,明显增高(P<0.05)。PTEN的表达下降与病理分级、肌层浸润程度、淋巴结转移有关(P<0.05);iNOS的过表达与病理分级、肌层浸润程度有关。子宫内膜癌组织中PTEN和iNOS的表达呈负相关。结论PTEN的表达下降或缺失与iNOS的过表达在子宫内膜癌的发生发展中可能起着一定的作用。 Objective To study the relationship between PTEN, inducible nitric oxide synthase ( iNOS ) protein expression and carcinogenesis and progression of endometrial carcinoma. Methods A specific and sensitive histochemical EnVisionassay was used to detect the expresion of PTEN and iNOS in paraffin embedded sections of 55 cases of endometrial carcinoma,24 atypical hyperplasia of endometrium and 20 normal endometrium. Results Compared with the positive expression rate of 100% in the normal endometrium, the expression of PTEN was reduced gradually in the hyperplasial endometrium and endometrial carcinoma( 91.67% and 60%, respectively) (P 〈 0.05) ; the positive rate of PTEN was strongly associated with pathological grade, muscles invasion and lymph node metastasis ( P 〈 0.05 ). The positive rate of iNOS plasm expression in carcinoma was significantly higher than that in normal endometrium and atypical hyperplasia of endometrium, which were 5.00% ,8.33% and 76.36% (P 〈 0.05). The positive rate of iNOS was strongly associated with pathological grade and muscles invasion ( P 〈 0.05 ). Conclusion The results show that low expression of PTEN and high expression of iNOS may relate with the development of endometrial carcinoma.
