摘要
以10%的扶正化瘀方药物血清作用于大鼠正常传代与纤维肝星状细胞、正常与纤维肝肝细胞、皮肤成纤维细胞,通过[3H-]-Propline掺入,观察药物血清对各组细胞胶原合成的影响,探讨扶正化瘀方抗肝纤维化的作用机制。结果发现,药物血清组纤维肝星状细胞与肝细胞细胞内胶原合成与细胞外胶原分泌均明显少于对照组;正常传代星状细胞与肝细胞细胞外胶原分泌也明显减少;成纤维细胞胶原的合成与分泌、以及正常传代星状细胞细胞内胶原合成也有抑制趋势。提示扶正化瘀方抗肝纤维化作用机制,可能主要是抑制了肝脏内胶原的合成。
To study the mechanism of Fuzhenghuayu (FZHY) decoction on liver fibrosis,10% FZHY decoction sera was incubated with rat normal subcultured cells and fibrotic liver stellate cells, normal and fibrotic hepatocytes and skill fibroblasts. The cells were impulsed with[3H-]-Propline, and cell collagen synthesis was measured. Our results showed that the intracellular collagen synthesis of fibrotic stellate cells and the hepatocytes in FZHY decoction group andtheir extracellular collagen secretion were both decreased obviously. Extracelluar collagen secretion of normal stellate cells and hepatocytes was decreased. The collagen production and secretionof fibroblasts, the intracellular collagen production of stellate cells was also inhibited to some extent. It is concluded that the mechanism of FZHY decoction on liver fibrosis may be associatedwith liver collagen production.
