苏氨酸对离体培养大鼠黄体细胞3β-羟-甾体脱氢酶活性的影响

摘要 用体外细胞培养的方法及放射免疫分析技术,观察L-苏氨酸对大鼠黄体细胞3β-羟-甾体脱氢酶(3β-HSD)活性的影响。结果显示,L-苏氨酸可明显抑制黄体细胞3β-HSD活性,而且在L-苏氨酸拮抗hCG致孕酮生成作用中,3β-HSD活性显著降低,提示L-苏氨酸可通过对3β-HSD活性的作用而影响卵巢黄体孕酮生成。 Previous studies in our laboratory showed that Lthreonine could not only inhibit hCGinduced and oxytocininduced progesterone production in the ovary in vitro,but also suppress progesterone secretion by human placental trophoblastic cells in vitro.In order to investigate the active mechanism of Lthreonine,we studied the effect of Lthreonine on 3βHSD activity of rat luteal cells in vitro by RIA.The results showed that 0.2mmol/L and 2.0mmol/L Lthreonine significantly inhibited 3betaHSD activity and hCG induced 3betaHSD activity.It suggests that 3βHSD might constitute an important site of action of Lthreonine during progesterone production affected by Lthreonine.
