摘要
目的探讨肝核受体(LXRs)激动剂T0901317对正常SD大鼠骨骼肌细胞中脂肪酸较运酶(FAT/CD36)基因mRNA表达的影响。方法将原代培养的5只SD大鼠骨骼肌细胞分为T0901317 1μmol/L作用组、0.5μmol/L作用组和未作用(阴性对照)组,采用SYBR Green I实时荧光定量聚合酶链反应(PCR)法检测各组SD大鼠骨骼肌细胞FAT/CD36基因mRNA表达水平,并进行比较分析。结果1μmol/L、0.5μmol/L作用组与阴性对照组比较,差异无显著性意义(P=0.116)。结论LXRs激动剂T0901317对SD大鼠骨骼肌细胞中FAT/CD36基因mRNA的表达水平没有明显作用,提示T0901317在促进SD大鼠骨骼肌细胞内脂肪酸的堆积作用尚无确切的证据。
Objective To investigate the Liver X receptors (LXRs) agonists T0901317' s effection on expression of FAT/CD36 gene mRNA in SD rat skeletal muscle cell. Methods Primary culture of Myotubes cells from SD rats were grouped to different T0901317 concentrations (0, 0. 5, and 1.0 μmol/1) for 24 hours before experiments. Then the expression of FAT/ CD36 gene mRNA in skeletal muscle cell of each group was detected by SYBR Green I real-time quantitative polymerase chain reaction. Results The Ct means of control group, T0901317 (0. 5μmol/l) group, T0901317 ( 1μmol/l) group were analyzed, and the difference has no significance meaning in three groups. Conclusion The expression of FAT/CD36 gene in SD rat skeletal muscle cell after the treatment of Liver X receptors agonists T0901317 was the same as control group, so we propose that T0901317 may not increase the risk of skeletal muscle resistance of SD rats.
