摘要
目的探讨幽门螺杆菌(Helicobacter pylori,Hp)感染及乳腺癌扩增1(amplified in breast cancer 1,AIB1)基因表达与胃癌的相关性。方法应用快速尿素酶法和改良吉姆萨(Giemsa)染色法检测Hp,用半定量逆转录-聚合酶链反应(RT-PCR)方法检测慢性浅表性胃炎组、中重度慢性萎缩性胃炎组及胃癌组中AIB1 mRNA的表达水平。结果胃癌组、中重度慢性萎缩性胃炎组和慢性浅表性胃炎组Hp感染率分别为73.3%、71.4%和43.3%,AIB1 mRNA表达水平分别为2.75±0.24、1.92±0.26和1.10±0.19。胃癌组及中重度慢性萎缩性胃炎组Hp感染率及AIB1 mRNA的表达水平都明显高于慢性浅表性胃炎组(P<0.05或<0.01)。中重度慢性萎缩性胃炎组及胃癌组Hp阳性AIB1 mRNA的表达水平明显高于Hp阴性(P<0.01)。结论Hp感染可能与胃黏膜组织中AIB1 mRNA的过表达有关,可能是其新的致癌机制之一。
Objective To explore the association among Helicobacter pylori（Hp） infection and expression of amplified in breast cancer 1 （AIB1） gene and gastric carcinoma. Methods Hp was assessed by using rapid urease staining test combined with modified Giemsa staining test. Semi-quantitative reverse transcription-polymerase chain reaction（RT-PCR） was used to detect the expression of AIB1mRNA in chronic superficial gastritis group and the modest and severe atrophic gastritis group and gastric carcinoma group. Results The infection rates of Hp in gastric carcinoma group, modest and severe atrophic gastritis group and chronic superficial gastritis group were 73.3%, 71.4% and 43.3%, respectively, and the expression of AIBlmRNA was 2.75 ± 0.24,1.92 ±0.26 and 1.10 ± 0.19, respectively. The infection rate of Hp was higher（ P 〈0.05） ,the expression of AIB1mRNA was higher（ P 〈0.01） in gastric carcinoma group and modest and severe atrophic gastritis group than that of chronic superficial gastritis group. The expression of AIB1mRNA in Hp positive group was higher than that in Hp negative group（ P 〈0.01）. Conclusion The infection of Hp might correlate with the over-expression of AIB1mRNA in gastric mucosa tissue,and it may be a new carcinogenesis mechanism.
