摘要
通过建立血清中甘氨胆酸浓度的测定方法,旨在为观察甘氨胆酸在大鼠血清中浓度的变化规律做试验依据,为开发有价值的动物源性药物做前期探索。试验采用固相萃取一反相高效液相色谱法(SPE—HPLC),色谱柱为Waters Xterra RP18(150mm×4.6mm.5μm),流动相为乙腈-KH2PO4溶液,检测波长213nm,柱温30℃,进样体积10μL。结果在0.15625~10.0μg/mL范围内甘氨胆酸浓度与峰面积呈良好的线性关系,回归方程Y=39579X+57823,R^2=0.9984;以信噪比S/N=3为标准,最低检测浓度为0.02μg/mL,平均回收率98.49%;日内RSD0.52%~2.00%,日间RSD0.74%~4.98%。该方法具有简便、稳定、灵敏、准确、实用等特点,适用于血清中甘氨胆酸含量的测定分析。
In this paper, determination method of concentrations of glycocholeic acid in rats blood serum is established by reversed-phase high performance liquid chromatography(RP-HPLC), This study aims to understand the variations of concentrations of glycocholeic acid in rats blood serum, provide a new method for Clinical Detection of glycocholeic acid, development useful drug from animal source. Uses the HPLC law, the chromatographic column is Waters Xterra RP18 ( 150 mm × 4.6 mm, 5 μm), flowing for second grade nitrile-KH2PO4 solution, examination wave length 213 nm, column warm 30℃, specimen handling volume 10 μL. The result the glycocholeic acid density and the peak area assumes the good linear relations in 0. 156 25 - 10.0 μg/mL scope, regression equation Y = 39 579 X + 57 823, R^2 = 0. 998 4 ; Take signal-to-noise ratio S/N = 3 as the standard, the lowest examination density is 0.02 μg/mL, The average recovery was 98.49% ;the RSD of inner-day was 0..52% - 2.00% ,the RSD of intra-day was 0.74% - 4.98% .The method was simple, accurate, sensitive, feasible and suitable for analyzing glycocholeic acid concentration in blood serum.
