摘要
采用酸水解法提取了重楼的水解产物薯蓣皂苷元,对其进行气相色谱分析,获得了相应的定量数据.气相色谱条件:HP-5MS熔融石英毛细管色谱柱(30.0m×320μm×0.25μm);FID检测器;载气流速2.5mL/min;进样口温度280℃;分流比10:1;检测器温度310℃;炉温270℃;进样量1.0μL.在最佳色谱条件下,薯蓣皂苷元在8.0~4000μg/mL范围内具有良好的线性关系,R^2=0.9991,测定方法平均回收率为89.0%,RSD为3.7%.GC测定结果和HPLC测定结果相近.
The diosgenin extracted by acid hydrolysis of dioscin was determined by gas chromatography. The sample was easily determined under the optimized conditions: chromatographic column (30.0 m× 320μm × 0.25/xm) ; FID detector; flow rate of carrier gas 2.5 mL/min; injecting temperature 280 ℃ ; split ratio 10:1 ; detector temperature 310℃ ; oven temperature 270℃. Method validation was conducted through using HPLC method to analyze the same sample to get quantitation result. The recovery and relative standard deviation (RSD) of the GC method was 89.0% and 3.7%, respectively. The linear calibration range was 8.0 -4 000 μg/mL for dioscin.
