96孔板法用于除草剂高通量筛选的研究 被引量:7

Application of 96-well plate method in screening herbicides at high throughput level

摘要 分别用三角瓶法和96孔板法研究了4种除草剂(精吡氟禾草灵、精喹禾灵、氯氟吡氧乙酸、乙草胺)对小球藻的72 h生长抑制急性毒性效应,结果表明:这四种除草剂均对小球藻表现出一定的水生毒性,用两种方法测得的除草剂对小球藻毒性趋势基本一致,半抑制浓度EC50值非常接近.96孔板法用于农药高通量筛选时,与国际标准组织推荐的三角瓶法的可靠性程度基本相同.三角瓶法筛选农药时需要毫克级的量,而96孔板法只需要微克级的量即可满足筛选的需要. The flask method and the 96-well plate method were used to investigate the acute toxicity of four kinds of herbicides (Fluazifop-P-butyl, Quizalofop-p-ethyl, Fluroxypyr, Acetochlor) to Chlorella vulgaris in this work. The results showed that all the herbicides had the aquatic toxicity to Chlorella vulgaris, the toxicity trend gained by two methods was mainly the same, and the value of EC50 was also contiguous. So the 96-well method had equal reliability to the flask method in screening herbicide at a high throughput level. Milligram grade of herbicide was needed in screening herbicide by the flask method, while it only took several microgrammes of herbicide by the 96-well plate method.
