摘要
[目的]探讨用铁氰化钾比色法测定烟叶总糖含量的影响因素。[方法]在测定试验中,设水浴时间4个水平(10、20、30、40 min),酸碱度4个水平(pH值为6.0、7.0、8.0、9.0),研究水浴时间和酸碱度对烟叶总糖测定的影响。[结果]烟叶总糖转化时水浴时间长短对测定结果影响较大,总糖转化后溶液pH值在一定范围内对测定结果无明显影响。同一样品水浴10 min与另外3个处理总糖的测量值差异极显著(P〈0.01)。总糖转化后将待测液pH值控制为6.0-9.0时,不同处理间所有样品总糖的测定值差异不显著(P〈0.05),但所有样品待测液pH值为9.0的总糖测量值均小于对应样品在pH值6.0、7.0、8.0时的测量值。[结论]烟叶总糖转化时间以80℃水浴10 min较为适宜,待测液pH值范围可控制为6.0-8.0较为适宜。
[Objective] The study aimed to discuss the factors influencing the determination of total sugar content in tobacco leaves by using ferricyanide colorimetric determination.[Method] In the determination test,the water-bath time at 4 levels of 10,20,30,40 min and the alkalinity acidity at 4 levels with pH of 6.0,7.0,8.0,9.0 were set up to study the effect of water-bath time and the alkalinity acidity on the determination of total sugar content in tobacco leaves.[Result] When the total sugar content in tobacco leaves was transformed,the water-bath time had greater effect on its determination results and after the transformation of total sugar the pH value in the transforming solution in a certain range had no significant impact on the determination results.The same sample in the treatment of water bath for 10 min had extremely significant difference with other 3 treatments on total sugar measurements(P〈0.01).When pH value in determined solution was controlled as 6.0-9.0 after the transformation of total sugar,the total sugar measurements of all samples among the different treatments had no significant difference(P〈0.05),but that of all samples measured at pH value of 9.0 were less than that of the corresponding samples measured at pH value of 6.0,7.0,8.0.[Conclusion] The transforming time for total sugar content in tobacco leaves was suitable to take water-bath time at 80 ℃ for 10 min and it was appropriate to control the pH value as 6.0-8.0 in determined solution.
