离子色谱-脉冲安培检测法测定酵母细胞壁中β-葡聚糖和甘露寡糖 被引量:7

Determination of β-Glucan and Mannan Oligosaccharides in the Yeast Cell Wall by Ion Chromatography and Pulsed Amperometric Detection

摘要 建立了高效阴离子交换色谱脉冲安培检测法分离测定酵母细胞壁中β-葡聚糖和甘露寡糖的方法。以NaOH为淋洗液,在CarboPac PA10阴离子交换柱上同时分离甘露糖醇、阿拉伯糖、半乳糖、果糖、葡萄糖和甘露糖。样品中的低聚糖在优化的水解条件下水解得到葡萄糖和甘露糖,通过测定2种单糖的含量而得到β-葡聚糖和甘露寡糖的含量。这2种单糖即对应低聚糖的检出限(25μL进样,S/N=3)分别为0.4和0.5pg/L,且均具有较宽的线性范围(0.002-50mg/L),3个样品测定的相对标准偏差在0.3%~1.0%。该方法检测糖简便快捷、分离效果好、无需衍生、灵敏度高,适用于多种复杂样品中的低聚糖分析。 A chromatographic method was developed for the determination of β--Glucan and Mannan Oli- gosaceharides in the yeast eell wall samples using High Performance Anion Exchange Chromatography coupled with Pulsed Amperometric Detection. Mannitol, arabinose, galactose, fructose, glucose and mannose can be separated well by CarboPac PA10 anion exchange column with sodium hydroxide elution. Oligosaccharides in these samples were hydrolyzed into glucose and mannose form with the optimized conditions, and the contents of β-Glucan and Mannan Oligosaccharides were calculated by the concentration of each monosaccharide. The detection limits of two oligosaecharides (25μL injection,S/N=3) as well as glucose and mannose , are 0.4μg/L and 0.5μg/L respectively. Moreover, the data have wide linearity range(0. 002-50 mg/L) and good relative standard deviation (0.3-1.0%). This method is suitable for the determination of oligo- saccharides in complex samples for its simple, fast and good separating performance as well as high sensitivity.
