摘要
目的:探讨全反式视黄酸(ATRA)对乳腺癌细胞生长抑制和对细胞凋亡诱导的机理。方法:采用荧光显微术、流式细胞术、MTT测定和RNA印迹等方法,检测细胞凋亡、细胞生长和mRNA的表达水平。结果:1)ATRA能够有效地诱导ZR-75-1乳腺癌细胞凋亡,但不能诱导MDA-MB-231乳腺癌细胞凋亡。2)ATRA能够有效地抑制ZR-75-1细胞的生长,但不能抑制MDA-MB-231细胞生长。3)ATRA能够诱导ZR-75-1细胞的视黄酸受体RARβ的表达,但不能诱导MDA-MB-231细胞的RARβ表达。结论:ATRA诱导的细胞凋亡与其对ZR-75-1细胞生长抑制有关,对MDA-MB-231细胞生长及凋亡无影响,RARβ的表达可能介导ATRA诱导的细胞凋亡和细胞生长抑制过程。
Objective: To explore the effect mechanism of all trans etinoic acid (ATRA) on induction of apoptosis and the growth inhibition in human breast cancer cell lines. Methods: Apoptosis, growth inhibition and mRNA level in breast cancer cell lines were detected by fluorescent microscopy, flow cytometry, MTT assay and northern blot. Results: 1) ATRA can induce apoptosis of ZR-75-1 breast cancer cells, but it can not induce apoptosis of MDA-MB-231 breast cancer cells. 2) ATRA can inhibit the growth of ZR-75-1 breast cancer cells effectively, whereas it has no effect on MDA-MB-231 breast cancer cells. 3) ATRA can induce RAR β expression in ZR-75-1 breast cancer cells, but it can not induce RAR β expression in MDA-MB-231 breast cancer cells. Conclusions: Induction of apoptosis may contribute to the growth inhibitory effect of ATRA in ZR-75-1 breast cancer cells. The expression of RAR β is an important factor which may mediate apoptosis induced by ATRA and growth inhibitory effect of ATRA.
