可催化生产生物柴油脂肪酶产生菌的筛选及其培养条件研究 被引量:10

Screening and culture condition study of strain producing lipase to catalyze the production of biodiesel

摘要 以筛选出的活性较高菌株Y7生产脂肪酶。此菌最佳产酶条件为:1.0%的葡萄糖和1.5%的NaHCO3作为C源,1.5%的蛋白胨作为N源,添加0.1%MgSO4.7H2O,初始pH9.0,30℃培养48h。在培养液中分别加入1.0%的不同天然油脂和有机酸进行研究,发现该菌对添加物有特异性,只有橄榄油有较强促进作用,而油酸、乳酸等具有较强的抑制作用。在添加1.0%的橄榄油时,产酶活力可达到66.31 U/mL。此酶最适作用温度为30℃,最适作用pH为8.0。 Strain Y7 screened with higher activity was used to produce lipase. The optimal culture conditions were determined as follows:1. 5% NaHCO3 and 1.0% glucose used as carbon sources,1.5% peptone used as nitrogen sources ,0.1% MgSO4 · 7H2O in culture medium ,initial pH 9.0 ,incubation at 30 ℃ for 48 h. It found that the strain had selective effect to the natural oils and organic acids, only olive oil had significant stimulation effect, while oleic acid or lactic acid could weaken the production of lipase greatly. Adding 1.0% olive oil to the culture medium, the enzyme activity reached 66.31 U/mL. The optimum reaction temperature and pH of the lipase obtained was 30 ℃ and 8.0, respectively.
