摘要
为了研究芎归胶艾汤的促孕保胎作用及其对母胎界面Th1/Th2型细胞因子的影响,取怀孕小鼠分成4组,分别为对照组、RU486组、RU486+孕酮组、RU486+芎归胶艾汤组。除对照组外,其它3组于孕4 d(GD4)7:00皮下注射RU486 0.5 mL,同时孕酮组注射0.1 mL孕酮,中药组灌服芎归胶艾汤0.8 mL,各组分别在GD4下午19:00,GD5上午7:00,GD5下午19:00取材。统计胚泡数,酶联免疫吸附法(ELISA)测定子宫匀浆中IFN-γ、IL-2、IL-4和IL-10的含量。结果表明:RU486显著降低胚泡数,Th1型细胞因子IFN-γ和IL-2的水平显著升高,Th2型细胞因子IL-4和IL-10的含量显著下降。补充孕酮或芎归胶艾汤显著逆转了RU486引起的流产,同时降低了Th1型细胞因子水平,提高了Th2型细胞因子水平,表明芎归胶艾汤能够作用在母胎界面,调节着床和妊娠期细胞因子平衡,利于胚胎着床和妊娠维持。
To investigate the effect of Xiong- gui- jiao-ai tang （XGJAT） on abortion model and its impact on Th1/Th2 cytokines at the maternal - fetal interface, the pregnant mice were divided into 4 groups： control group, RU486 group, RU486 ＋ progesterone （P） group and RU486 ＋ XG- JAT group. All mice except the control were injected with 0.5 mL RU486 subcutaneously on day 4 of pregnancy to establish abortion model. At the same time, the mice in RU486 ＋ P group were in-jetted with 0.1 mL progesterone and those of RU486 ＋ XGJAT group gavaged with 0.8 mL Xiong -gui-jiao-ai tang. Samples were collected in 19：00 on day 4, 7：00 and 19：00 on day 5. The number of embryos were calculated and the contents of IFN -γ, IL- 2, IL - 4 and IL - 10 in uterine supernatant were measured by Enzyme- Linked Immuno- absorbent Assay （ELISA）. Results showed that, with decreased number of embryos, the contents of IFN -γ and IL- 2 which represent Thl cytokines significantly increased, while Thl cytokines, such as IL- 4 and IL- 10, significantly decreased after treatment with RU486. Supplement with progesterone or Xiong - gui - jiao- ai tang efficiently attenuated the abortive effect of RU486 through the decreasing of Thl cytokines and increasing the Th2 cytokines, indicating that Xiong- gui- jiao- ai tang modulates cytokine network in pregnancy and benefits embryo implantation and survival.
